The ion channels activated by glutamate are typically divided into two classes. Glutamate receptors that are activated by kainate and alpha-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid (AMPA) are known as kainate/AMPA receptors (K/AMPAR). Those that are sensitive to N-methyl-D-aspartate (NMDA) are designated NMDA receptors (NMDAR). The NMDAR plays an essential role in memory, neuronal development and it has also been implicated in several disorders of the central nervous system including Alzheimer's, epilepsy and ischemic neuronal cell death. The rat NMDAR1 (NR1) was the first subunit of the NMDAR to be cloned. The NR1 protein can form NMDA activated channels when expressed in Xenopus oocytes but the currents in such channels are much smaller than those seen in situ. Channels with more physiological characteristics are produced when the NR1 subunit is combined with one or more of the NMDAR2 (NR2 A-D) subunits. The NR2B subunit of the NMDARs is phosphorylated by cyclin dependent kinase-5 (cdk5) at Ser1252 in vitro and in intact cells and Ser1252 phosphorylation upregulates NMDA receptor channel activity. The phosphorylation at this site may also play a key role in ischemic cell death. (1-6)\n\nApplications: \nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Specific for ~180k NMDAR NR2B subunit protein phosphorylated at Tyr1336. Immunolabeling of the NMDAR NR2B subunit band is blocked by l-phosphatase treatment. Species Crossreactivity: Human, mouse and rat based on 100% antigen sequence homology.
