CARD15 (Nod2) is a member of the Nod1/Apaf-1 family of intracellular proteins that contain an N-terminal caspase recruitment domain (CARD), a centrally located nucleotide-binding domain (NBD), and a C-terminal regulatory domain. CARD15 induces NF-kappa-B via RICK (CARDIAK, RIP2) and IKK-gamma. It also confers responsiveness to intracellular bacterial lipopolysaccharides (LPS). Unlike Nod1, this homologue is primary expressed in monocytes. Defects in CARD15 are associated with Blau Syndrome (BS) and Crohn's Disease (CD), and are a cause of susceptibility to ulcerative colitis.\n\nPositive Control: Human Peripheral Blood Mononucleocyte\n\nApplications: \nSuitable for use in ELISA and Western Blot. Other applications not tested.\n\nRecommended Dilution:\nELISA: 1:64,000.\nWestern Blot: 0.1-0.3ug/ml. ~100kD band observed in lysates of Human Peripheral Blood Mononucleocytes (calculated MW of 115kD according to NP_071445.1).\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\n\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
