Endothelial nitric-oxide synthase (eNOS is an important enzyme in the cardiovascular system. It catalyzes the production of nitric oxide (NO), a key regulator of blood pressure, vascular remodeling and angiogenesis (1,2). eNOS is regulated by phosphorylation at multiple sites. The two most thoroughly studied sites are the activation site serine 1177 and the inhibitory\nsite threonine 495 (3). Several protein kinases including Akt/PKB, PKA and AMPK activate eNOS by phosphorylating serine 1177 in response to various stimuli (4,5). In contrast, bradykinin and hydrogen peroxide activate eNOS activity by promoting threonine 495 dephosphorylation (6,7). eNOS is activated by VEGF, and this activation is associated with dephosphorylation of eNOS at serine 113. Cyclosporin A blocks this dephosphorylation of eNOS upon VEGF stimulation (8).\n\nApplications: \nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:1000\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot. Store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
