NT-3 is a neurotrophic factor structurally related to NGF, BDNF, and NT-4. These proteins belong to the cysteine-knot family of growth factors that assume stable dimeric structures. NT-3 is expressed by neurons of the central nervous systems and can signal through the trk receptors. NT-3 promotes the growth and survival of nerve and glial cells. The amino acid sequences of human, murine and rat NT-3 are identical.\n\nDescription:\nRecombinant Human NT-3 is a single, non-glycosylated polypeptide chain containing 119 amino acids and having a molecular mass of 13606.29 Dalton.\n\nSequence:\nThe sequence of the first five N-terminal amino acids was determined and was found to be Tyr-Ala-Glu-His-Lys.\n\nBiological Activity:\nThe ED50 as determined by the dose-dependent induction of choline acetyl transferase in rat basal forebrain primary septal culture was found between 20-50ng/ml.\n\nProtein Content:\nProtein quantitation was carried out by two independent methods:\n1. UV spectroscopy at 280 nm using the absorbency value of 2.165 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). \n2. Analysis by RP-HPLC, using a standard solution of NT-3 as a Reference Standard.\n\nStorage and Stability:\nLyophilized powder may be stored at 4 degrees C for short-term only. Reconstitute to nominal volume by adding sterile dH2O and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
