p130 Cas (Cas3)

Many aspects of signal transduction are regulated by protein-protein interactions and involve a class of molecules called adapter or scaffolding proteins. p130 CAS is a newly described member of this group of proteins that contains numerous sequence motifs predicted to be involved in protein-protein interactions. p130 CAS was first identified as a protein involved in src-mediated transformation, but it is now thought to play a role in many other signaling pathways as well, including cellular transformation by the v-crk oncogene, mitogenesis, and adhesion. \n\nApplications:\nSuitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested.\n\nRecommended Dilution:\nWestern Blot: 0.5-2ug/ml detects p130 CAS in RIPA lysates from human 3T3/A431 cells. A previous lot detected p130 Cas in RIPA lysates from human A431, HFF, mouse 3T3 and rat L929 cells. 3T3/A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with P1001-97 (0.5ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.\nImmunoprecipitation: 4ug immunoprecipitates p130 CAS from 500ug of 3T3 RIPA lysate.\nOptimal dilutions to be determined by the researcher.\n\nPositive Antigen Control (Included): \nF4209 Fibroblast Cell, 3T3/A31: Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
