C-KDFPEEDDPTN from the internal region of human NCF4.\nControl peptide corresponding to polyclonal antibody, Catalog#P1001-20G, goat x human.\n\nThe NADPH oxidase generates superoxide in phagocytic cells. It is important for immunity and its deficiency leads to chronic granulomatous disease (CGD). p40-phox is a newly isolated cytosolic component of the nicotinamide adenine dinucleotide phosphate (NADPH)-oxidase that copurifies with p67-phox. Preliminary evidence indicates that it is a component of the cytosolic complex. Maximal activation of NADPH oxidase requires formation of a complex between the p40-phox and p67-phox subunits via association of their PB1 domains. The crystal structure of the p40-phox/p67phox PB1 heterodimer reveals that both domains have a beta grasp topology and that they bind in a front-to-back arrangement through conserved electrostatic interactions between an acidic OPCA motif on p40-phox and basic residues in p67-phox.\n\nApplications:\nSuitable for use in ELISA. Other applications not tested.\n\nRecommended Dilutions:\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 monthsat -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.\n\nPeptide Blocking (see corresponding antibody P1001-20G):\nAntibodies are typically supplied at 0.5mg/ml and peptides as a 100ul pellet. When peptides are reconstituted in 200ul water, the concentration would be also 0.5mg/ml. To start, the best ratio would be 1:1 (which means molar excess of peptides relative to antibodies when identical volumes are mixed). Mix equal volumes of peptide and antibody at the required dilution and leave at ambient temperature. It is best is to have two identical blots, to be incubated with equal amount of antibodies, but one with the antibodies pre-adsorbed to the peptide for 20min. Then incubate and develop the two blots in parallel.
