Parvalbumin, Recombinant, Rat

Parvalbumin (PV or PVALB or oncomodulin) is a high affinity low mol wt (~12kD) cytoplasmic protein belonging to the troponin C superfamily. It is expressed in high levels only in fast-contracting muscles and at lower levels in brain and several endocrine tissues. PV has two isoform: alpha (rat/mouse/human 110-aa, 7p13-p11) beta form (mouse/rat/human 109-aa) shares approx. 48% identity (mouse PVs). The PV-alpha is less acidic (pI=5) than the Beta-isoform and has an additional amino acid in the C-terminal helix. PV-alpha is also expressed in inner ear. PV-beta isoform is expressed in many fetal tissues, placenta and tumors.\n\nApplications: \nThe Parvalbumin can be used for immunoblots, absorption experiments in immunohistochemistry, radioimmunoassay and intracellular injection. For fluid-phase adsorption in immunohistochemistry we suggest the following procedure: A- Dilute 1 microl of the mon oclonal antibody 235 (Swant) against parvalbumin in 5 ml of the usual buffer for immunohistochemistry (final dilution 1:5000). B- Add 1microg of the Parvalbumin to 1ml of the diluted antibody solution and mix well. C- Incubate for at least 6 hours in the cold. D- Apply to tissue-sections and incubate for 3 days. E- Complete the immunohistochemical reaction as usual (biotinylated second antibody, ABC-complex,DAB). As a result, the immunostaining should be strongly reduced or even completely prevented.\n\nSolubility: \nIt is recommended to reconstitute the lyophilized Parvalbumin in sterile 18MOmega-cm H2O not less than 100microg/ml, which can then be further diluted to other aqueous solutions.\n\nStability: \nLyophilized Parvalbumin although stable at room temperature for 3 weeks, should be stored desiccated below -18 degrees C. Upon reconstitution Parvalbumin should be stored at 4 degrees C between 2-7 days and for future use below --18 degrees C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.
