Phosphotyrosine

Protein tyrosine phosphorylation plays a central role in the transmission of signals from the cell surface to the nucleus. Reversible phosphorylation of cellular proteins on specific tyrosine residues is involved in a variety of fundamental cellular processes including survival, proliferation, differentiation, and transformation. Moreover, the use of antibodies which specifically recognize phosphotyrosine is widely accepted as one of the most powerful tools for detection, characterization and purification of proteins which are either constitutively or inducibly phosphorylated on tyrosine.\n\nApplications: \nSuitable for use in Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1ug/ml\nImmunoprecipitation: 5ug\nInhibition: Can be achieved by pre-incubation of the antibody with 20mM phosphotyrosine, whereas pre-incubation with 20mM phosphoserine or 20mM phosphothreonine is ineffective.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Reacts specifically with native and denatured tyrosine-phosphorylated proteins. It shows no crossreactivity to phosphoserine or phosphothreonine. Positive reactivity is independent of the species and has been observed in the following samples: mouse brain, NIH 3T3 cells (+/-TPA), K562 cells, EGF-stimulated A431 cells and MCF-7cells.
