Platelet Derived Growth Factor BB, Recombinant, Mouse (PDGF-BB)

Recombinant Mouse PDGF-BB is a homodimeric, non-glycosylated, polypeptide chain containing 109 amino acids and having a molecular mass of 25kD. \n\nPDGF is a mitogenic peptide growth hormone carried in the alpha-granules of platelets. It is released when platelets adhere to traumatized tissues. Connective tissue cells near the traumatized region respond by initiating the process of replication.The synthesis of PDGF can be induced by Interleukin-1, Interleukin-6, TNF-alpha, TGF-beta and EGF.\n\nAmino Acid Sequence: \nThe sequence of the first five N-terminal amino acids was determined and was found to be Met-Ser-Leu-Gly-Ser.\n\nBiological Activity: \nThe biological activity is determined by the dose-dependant stimulation of the proliferation of human umbelical vein endothelial cells (HUVEC) using a concentration range of less than 1.6ng/ml.\n\nProtein Content: \nProtein quantitation was carried out by two independent methods:\n1. UV spectroscopy at 280nm using an extinction coefficient of 0.48 for a 0.1% solution\n2. Analysis by RP-HPLC, using a standard solution of PDGF-BB as a reference standard\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile 100mM acetic acid, 0.1% BSA. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
