RAD50, a member of the RAD52 epistasis group, is believed to play an important role in the nucleolytic processing of DNA double-stranded breaks. Studies in Saccharomyces cerevisiae show that mutation in the RAD52 family of proteins causes varying degrees of mitotic and meiotic recombination, both of which are initiated via DNA double-stranded breakage. Rad50 forms a complex with Mre11, another RAD52 family member, and p95 Nibrin, a product of the mutated gene associated with Nijmegen breakage syndrome. This complex possesses both endonuclease activity and a 3' and 5' exonuclease activity. RAD50 is indispensable for non-homologous DNA end joining and the maintenance of telomere length. \n\nImmunoblot Analysis: 0.5-2ug/ml detects RAD50 in a HeLa nuclear lysate. HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Rad50 (0.5ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. \n\nPositive Antigen Control: HeLa nuclear lysate. Use 20ug per lane for minigels.\n\nImmunoprecipitation: Reported to immunoprecipitate. \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C or colder. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
