The Ras pathway is a critical signal transduction cascade involved in regulating cellular proliferation, differentiation, survival, and oncogenic transformation. Members of the Raf serine/threonine kinase family are key intermediates in this cascade, functioning to relay signals from activated Ras to the downstream protein kinases MEK and ERK. Previous studies have shown that phosphorylation is required for Raf-1 activation. Recent work has demonstrated that phosphorylation also regulates the downregulation of Raf with two sites participating Ser 301 and Ser 642.\n\nApplications: \nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 3 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Specific for the ~74k Raf-1 protein phosphorylated at Ser642. Immunolabeling of the Raf-1 protein band is blocked by l-phosphatase treatment. Species Crossreactivity: Human, mouse, and rat based on 100% antigen sequence homology.
