Renin is an important enzyme in the regulation of blood pressure and electrolyte homeostasis and therefore is of great clinical importance. For these same reasons, the kinetic and mechanistic functions of the enzyme are of importance for the development of hypertension drugs. Renin, also known as angiotensinogenase, is a circulating enzyme released mainly by juxtaglomerular cells in the JGA of the kidneys in response to low blood volume or low body NaCl content, mediated through the release of prostaglandins. Although it has hormone-like actions, it cleaves a protein precursor in the circulation rather than working on a cellular target.\n\nMolecular Weight: ~40,000D\n\nSpecific Activity:\n~100pmoles of substr. hydrolyzed/min/ug\n\nActivity Measurement:\nActivity was measured by the cleavage of a fluorescent peptide substrate (Arg-Glu(EDANS)-Ile-His-Pro-Phe-His-Leu- Val-Ile-His-Thr-Lys(dabcyl)-Arg) and activity was detected in a microplate format using a SpectroMax Gemini microplate reader. \n\nStorage and Stability:\nFor long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
