Retinoblastoma, phosphorylated (Ser608) (Rb)

Entry into S phase from late G1 is controlled by phosphorylation of the retinoblastoma protein (Rb). Mutations affecting Rb or proteins that control Rb phosphorylation are frequent in human tumors. The cell cycle distribution of childhood acute lymphoblastic leukemia (ALL) cells shares many features with that of normal CD34+ progenitor cells. The majority of CD34+ and ALL cells reside in G1 phase although their cell cycle distribution within the G1 phase may be different. Entry of the cell into late G1 phase is controlled by phosphorylation of Rb. Different phosphoforms of Rb were recognized by their slower mobility on SDS-PAGE in ALL cells vs. CD34+ cells. In ALL cells, Rb was phosphorylated at ser-608, ser-780 and ser-795 unlike CD34+ cells. This evidence suggests that G1 phase distribution of ALL and CD34+ cells is different and that ALL cells progress to mid G1 phase while unstimulated CD34+ cells reside in early G1 phase. \n\nApplications: \nSuitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilution: \nELISA: 0.1-1ug/ml\nWestern Blot: .1-5ug/ml \nImmunoprecipitation: 10ug/IP reaction \nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nFor long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
