Retinoblastoma (Rb)

Retinoblastoma (Rb) is expressed in most normal cells and acts as a tumor suppressor gene product. Rb interacts with several cell cycle proteins including cyclin A, cyclin D, cyclin E and E2F transcription factors. Recent evidence shows that Rb protein is cleaved by caspases and a caspase-resistant Rb can attenuate the death response to tumor necrosis factor alpha, suggesting that Rb degradation contributes to the activation of cell-death pathway.\n\nApplications: \nSuitable for use in Immunofluorescence, Flow Cytometry, ELISA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1-2ug/ml for 2hrs at RT.\nImmunoprecipitation: 20ul/mg of cell lysate\nImmunohistochemistry (Frozen or formalin-fixed tissues): 1:50-1:100; Formalin-fixed tissue sections require boiling of sections in 1mM EDTA, pH 8.0, for 10-20 minutes followed by cooling at RT for 20 minutes.\nOptimal dilutions to be determined by the researcher.\n\nRecommended Positive Control:\nLS174T, HT29, HUVEC, ML-1, U2OS or Molt-4 cells and normal colon.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes a nuclear phosphoprotein of 105kD which is identified as the retinoblastoma (Rb) gene product. Its epitope is localized between aa703-772. It reacts with hyperphosphorylated Rb as well as underphosphorylated form of the Rb protein and shows no crossreaction with other members of Rb family such as p107 or p130. Species Crossreactivity: Human; Does not react with mouse or rat.
