Rb is a tumor suppressor nuclear phosphoprotein capable of binding to DNA. It is phosphorylated on serine and threonine, but not on tyrosine residues. It forms a complex with SV40 large T antigen, adenovirus E1A, and human papilloma virus-16 E. Rb protein may act by regulating transcription and loss of ts function leads to uncontrolled cell growth. Aberrations in the RB gene have been implicated in cancers of breast, colon, prostate, kidney, nasopharynx, and leukemia.\n\nApplications: \nSuitable for use in RT, Immunofluorescence, Flow Cytometry, Western Blot, Immunoprecipitation and Immunohistochemistry (paraffin-embedded sections). Other applications not tested.\n\nRecommended Dilution:\nOptimal dilutions to be determined by the researcher.\n\nCellular Localization: Nuclear\n\nPositive Control: Ls174T, HT29, HUVEC cells. Normal colon and colonic / breast carcinoma tissue sections.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 3 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional Info

Specifically stains the nuclei of BT-20 cells and primary human foreskin fibroblast (HFF) cells. Reacts with hyperphosphorylated and un (under) phosphorylated forms of Rb protein. It shows no crossreaction with p107 and p130. Species Crossreactivity: Crossreacts with Human. Does not crossreact with Mouse or Rat.
