Retinoic Acid Receptor Responder 1 (RARRES1, Tig1)

RARRES1 (Retinoic Acid Receptor Responder 1; also Tig1) is a member of the protease inhibitor I47 family of proteins. It is expressed in epithelium, including keratinocytes, plus prostate and breast epithelium. It is considered to be a tumor-suppressor molecule, presumably acting as an inhibitor of carboxypeptidase A. Mouse RARRES1 is 349 amino acids (aa) in length. Based on human, it is a type II transmembrane (TM) protein with a TM segment at, or near, aa75-95. One potential 350aa splice variant exists that shows an almost complete aa divergence after Pro105.\n\nApplications: \nSuitable for use in Direct ELISA, Western Blot and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nDirect ELISA: 0.5-1ug/ml; Detection limit is ~0.3ng/well.\nWestern Blot: 1ug/ml\nImmunohistochemistry (Fluorescence): 10ug/ml; Detects RARRES1 in cryosections of mouse liver. Tissue was fixed overnight in 4% PFA + 14% picric acid in PBS at 4 degrees C. Sections were blocked with 10% donkey serum and 0.1% Triton X-100 in PBS, 1% BSA for 45 minutes at RT. After blocking, sections were incubated with R1691-97 for 3 hours at RT, followed by donkey anti-goat IgG (NL557) for 1 hour at RT. Between each step, cells were washed with PBS, 0.1% BSA.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
