RICK (RIP2, CARDIAK, RIP-like Interacting CLARP Kinase, Receptor Interacting Protein-2, CARD-containing ICE Associated Kinase)

Apoptosis can be regulated by death domain (DD) and/or caspase recruitment domain (CARD) containing adapter molecules and a caspase (ICE/CED-3) family of cysteine proteases. DD-containing serine/threonine kinase RIP is an adapter molecule that mediates divergent signaling pathways including NF-kappaB activation and Fas (CD95) induced apoptosis. Recently identified, was a novel CARD containing serine/threonine kinase that regulates apoptosis which was designated RICK (RIP-like interacting CLARP [also known as FLIP] kinase) (1). RICK is also known as RIP2 (receptor interacting protein 2) and CARDIAK (CARD-containing ICE associated kinase) (2,3). RICK contains an N-terminal kinase catalytic domain and a C-terminal CARD domain. RICK kinase domain has high sequence homology to that of RIP. Overexpression of RICK promoted the activation of caspase-8 and Fas induced-apoptosis. RICK represents a novel kinase that regulates Fas induced-apoptosis. RICK mRNA is expressed in multiple human tissues (1). \n\nApplications:\nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot (ECL): 0.5ug/ml \nOptimal dilutions to be determined by researcher.\n \nPositive Controls:\nHeLa Cell Lysate\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20 degrees C or colder. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional Info

Detects a 60kD protein, corresponding to the apparent molecular mass of RICK on SDS-PAGE immunoblots, in samples from human, mouse, rat and bovine. Additional bands at ~130kD and 50kD are also observed on immunoblots. The antibody specificity is confirmed by peptide inhibition immunoblotting studies.
