RNP AO, phosphorylated (Ser84) (Nuclear Ribonucleoprotein, nRNP, hnRNP, Heterogeneous Nuclear Ribonucleoprotein A0)

Heterogeneous nuclear ribonucleoproteins (hnRNP) bind to nascent RNA polymerase II transcripts and are involved in transcript-specific packaging as well as alternative splicing of pre-mRNAs. hnRNPs also appear to influence mRNA metabolism and transport. In vivo, the M proteins bind to mRNA, and in vitro they bind avidly to poly(G) and poly(U) homopolymers. Four different isoforms of hnRNP are produced by alternative splicing: M1, M2, M3, and M4. Ongoing studies have theorized that hnRNPs may cause certain genetic diseases such as myotonic dystrophy, which is the most common form of adult muscular dystrophy. The identification of hnRNPs may therefore be important to determining the cause and cure for genetic diseases.\n\nApplications: \nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 0.5-2ug/ml detects phosphorylated hnRNP A0 in RIPA lysates from RAW 264.7 cells stimulated with LPS and recombinant hnRNP A0 that had been phosphorylated with MAPKAP K2. GST-hnRNP A0 was phosphorylated by MAPKAP K2, resolved by electrophoresis, transferred to nitrocellulose and probed with anti-hnRNP A0 (2ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.\n\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
