Members of the RSK (the p90 ribosomal S6 kinase) family of serine/threonine kinases are substrates of ERK (the Ras/extracellular signal-regulated kinase). The RSK family consists of four isoforms (RSK1 to -4) and two structurally related cousins, called RSK-like protein kinase (RLPK/MSK1) and RSKB (MSK2). RSKs have diverse functions, such as regulation of glycogen metabolism, cell survival of cerebellar neurons, and the regulation of transcription factors and coregulators, such as CREB, CREB-binding protein, p300, c-Fos, and estrogen receptor. RSK3 translocates to the cell nucleus, phosphorylates potential nuclear targets, and may have a unique upstream activator. RSK3 transcripts are present in most tissues, including heart, brain, placenta, liver, kidney and pancreas, with predominant expression in skeletal muscle.\n\nApplications:\nSuitable for use in Western Blot. Other applications have not been tested.\n\nRecommended Dilution:\nWestern Blot: 1-3ug/ml\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Reacts with mouse RSK3, and does not react with overexpressed RSK1, RSK2 or RSK4. On Western blots, it identifies a single band at ~83kD. Reactivity has been confirmed with 2 day-old mouse heart, fetal mouse muscle homogenates, and RSK3-transfected HEK293 cells.
