Ryanodine Receptor

Ryanodine receptors have been shown to play critical roles in the intracellular Ca2+ signaling occurring during cell activation in muscle cells and non muscle cells. \n\nApplications:\nSuitable for use in Immunoblot, Immunohistochemistry, Immunocytochemistry, Immunofluorescence, ELISA, Immunoprecipitation, Flow Cytometry and RIA. Other applications not tested.\n\nRecommended Dilution:\nImmunoblot (Enhanced Chemiluminescence Detection): 1:100. Perform a dot blot to verify presence of the RyR protein if having difficulties performing an immunoblot assay.\nImmunohistochemistry: 1:100\nImmunocytochemistry (Indirect Immunofluorescence): 1:100 Visualization by confocal microscopy is required, as detection by standard fluorescent microscopy will not be adequate to detect the RyR. Due to the intensity of confocal lasers, use of an anti-fading agent, such as DABCO, is strongly recommended. Additionally, fluorescent, not enzymatic, detection is required.\nELISA: 1:1000\nImmunoprecipitation: 1:100\nFlow Cytometry: 1:500\nRIA: 1:5000\n\nOptimal dilutions to be determined by the researcher.\n\nSource: \nTissue culture supernatant\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
