S100A1

S100 belongs to the family of calcium binding proteins such as calmodulin and troponin C. S100A is composed of an alpha and beta chain whereas S100B is composed of two beta chains. S100 protein is also expressed in the antigen presenting cells such as the Langerhans cells in skin and interdigitating reticulum cells in the paracortex of lymph nodes. S100 is a nervous tissue specific soluble protein made in glial cells. It is present in sweat glands chongrocytes and myoepithelial cells, and may be found as cytoplasmic, membranous, or mixed throughout the areas of tumor (usually cytoplasmic when present in normal tissues). S100 Protein derived from brain tissue is an acidic calcium-binding protein with the molecular weight of ~21kD. In human brain tissue S100 protein is mainly presented by two isoforms, beta-beta homodimer (S100b) and alpha-beta heterodimer (S100a).\nAntibody to S100 stains Schwannomas, ependymomas, astrogliomas, almost all benign and malignant melanomas and their metastases. \n\nApplications:\nSuitable for use in Immunohistochemistry, Immunocytochemistry, Indirect Immunofluorescence, Radioimmunoassay and Western Blot. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 1:50 using an avidin-biotin secondary antibody detection.\nIndirect Immunofluorescence: 1:2\nOptimal dilution to be determined by researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Reacts with bovine and porcine S100A1 in Immunoblots. Immunoreactivity with recombinant rat and human S100A1 and recombinant human S100A4 is reduced when compared to native bovine and porcine S100A1. Does not react with human S100A2 in immunoblots. Reacts with bovine, rat, and recombinant S100B in immunoblots. Less than 5% crossreactivity with S100B as seen in competition radioimmunoassays. Crossreactivity with other members of the S100 family has not been investigated.
