Stress-activated Protein Kinases (SAPKs) are strongly activated in response to adverse stimuli such as heat and osmotic shock, UV light and other DNA-damaging reagents, and inhibitors of protein synthesis. They are also activated strongly in response to agonists that are released or produced under conditions of stress, such as proinflammatory cytokines. Stress-activated Protein Kinase 2 (SAPK2/p38) phosphorylates conserved Ser-Pro or Thr-Pro peptide motifs.\n\nApplications: \nSuitable for use in ELISA and Western Blot. Other applications not tested.\n\nRecommended Dilution:\nELISA: 0.05ug/ml\nWestern Blot: 1ug/ml (Alkphos detection), 0.1ug/ml (HRP/ECL detection).\nOptimal dilutions to be determined by the researcher.\n\nPositive Control (included): \nA0001-50B A431 cell lysate, untreated\nAdd 75ul 2x SDS-PAGE sample buffer, mix and incubate at 90 degrees C for 5 min.\nWestern Blot: 5ul/lane (mini gel) for HRP/ECL detection; 20ul/lane (mini gel) for HRP/DAB detection.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
