SAPKa (SAPK-alpha, SAPK, c-Jun N-terminal Kinase 2, JNK2, Mitogen-activated Protein Kinase 9, MAP Kinase 9, MAPK 9, MAPK9, PRKM9, p54-alpha, Stress-activated Protein Kinase JNK2)

The stress-activated protein kinase alpha (SAPKalpha), also know as c-Jun NH2-terminal kinase 2 (JNK2) is a 48kD serine/threonine protein kinase with an apparent molecular masses on SDS-PAGE of 46 and 55kD due to alternative splicing. SAPKalpha is a member of the MAP kinase family. It is activated by cellular stress, tumor necrosis factor-alpha and sphingomyelin-based second messengers. The extracellular stimuli leading to cell stress include inflammatory cytokines (via Rho and Rac) or light, some growth factors, protein synthesis inhibitors and osmotic stress. SAPKalpha binds to the N-terminal activation domain of c-Jun and phosphorylates the regulatory sites Ser-63 and Ser-73. Two isoforms, alpha-2 and alpha-1 are produced as a result of alternative splicing.\n \nApplications: \nSuitable for use in Western Blot and ELISA. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot (Colorimetric): 1:500 Detes of SAPKa in 20ug of rat brain tissue extract. Recommend using a goat anti-rabbit IgG labeled with AP as detection antibody.\nOptimal dilutions to be determined by researcher.\n \nPositive Controls:\nRat Brain Tissue Extract\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add glycerol (40-50%) and aliquot. Store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes a 55kD protein, corresponding to the apparent molecular masses of SAPKalpha of rat and mouse on SDS-PAGE immunoblots. In human cell lysates, a 46kD band is also observed. Due to alternative splicing of the 46 and 55kD transcripts, numerous antigen specific bands are seen within this molecular mass range. A recombinant SAPKalpha preparation is strongly detected on immunoblots. Species Crossreactivity: Rat, mouse, human.
