A subset of the tumor necrosis factor (TNF) receptor family contain a conserved intracellular motif, the death domain. A member of this family, TRICK2, the TRAIL (TNF-related apoptosis-inducing ligand) receptor inducer of cell killing 2) is expressed in a number of cell types, and to particularly high levels in lymphocytes and spleen. TRICK2, like DR4, is a receptor for TRAIL:APO-2L [2,3] and could inhibit TRAIL-induced killing.\n\nApplications:\nSuitable for use in ELISA and Western Blot. Other applications not tested.\n\nRecommended Dilution:\nELISA: To detect hsTRAIL Receptor-2 by sandwich ELISA (using 100ul/well antibody solution) a concentration of 0.25-1ug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with Polyclonal Anti-Human sTRAIL Receptor-2 as a capture antibody, allows the detection of at least 0.2-0.4ng/well of recombinant hsTRAIL Receptor-2.\nELISA (Direct): To detect hsTRAIL Receptor-2 by direct ELISA (using 100ul/well antibody solution) a concentration of 0.25-1ug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with compatible secondary reagents, allows the detection of at least 0.2-0.4ng/well of recombinant hsTRAIL Receptor-2.\nWestern Blot: To detect hsTRAIL Receptor-2 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hsTRAIL Receptor-2 is 1.5-3ng/lane, under either reducing or non-reducing conditions.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile dH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
