Sec61b (Sec61 beta subunit, Protein transport protein Sec61 subunit beta)

The SEC61 complex is the central component of the protein translocation apparatus of the endoplasmic reticulum (ER) membrane. Oligomers of the SEC61 complex form a transmembrane channel where proteins are translocated across and integrated into the ER membrane. This complex consists of three membrane proteins alpha, beta, and gamma. This gene encodes the beta subunit protein. The SEC61 subunits are also observed in the post ER compartment, suggesting that these proteins can escape the ER and recycle back. There is evidence for multiple polyadenylated sites for this transcript.\n\nApplications:\nSuitable for use in Western Blot and Immunopurification. Other applications not tested. \n\nRecommended Dilutions: \nWestern Blot: A 1:10,000 dilution detects Sec61b in RIPA lysates from non-stimulated A431 cells. A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with S0580-10. Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system. \nOptimal dilution determined by the researcher. \n\nRecommended Secondary Antibodies:\nI1904-39: IgG, X-Adsorbed (HRP) Pab Gt x Rb\nI1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt x Rb\nI1904-46Q: IgG, H&L (HRP) Pab Gt x Rb\n\nPositive Antigen Control:\nA0001-50: Non-stimulated A431 cell lysate. Add 2.5ul of \n2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
