DEC-2 is a 54-56kD Class B member of the bHLH superfamily of proteins. It utilizes both DNA binding and protein-protein interaction to mediate E-box transcriptional repression. Physiologically, DEC-2 is proposed to be a regulator of the sleep cycle, particularly during periods of sleep deprivation. It also interacts with HIF-1 alpha neutralizing its activation of the VEGF gene during hypoxia. Human DEC-2 is 482aa in length. It contains a DNA binding motif aa45-57, an HLH region aa58-100, an Orange domain that may impart specificity to protein-protein interactions aa131-166 and a Gly/Ala-rich region aa297-431. DEC-2 forms homodimers, and heterodimers with MyoD and E-proteins. There is one potential alternate start site at Met280. Over aa101-287, there is less than 60% aa identity between human and mouse DEC-2.\n\nApplications:\nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1ug/ml\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
