Smad1 (MADR1, Small Mothers Against Decapentaplegic Deleted in Pancreatic Carcinoma)

The MAD-related (MADR) family of proteins are intracellular signaling components for the TGFb superfamily of ligands. MADR1/Smad1 is the mammalian homologue of the Drosophila protein MAD. MADR1/Smad1 has been shown to mediate BMP signals in the cell. It is phosphorylated on serine residues at the C-terminus and forms heterodimers with another MAD family member, DPC4 in response to BMP2 and BMP4. In addition, MADR proteins that are distributed throughout the cell become localized to the nucleus following its activation. These proteins can act as transcriptional activators when fused to a heterologous DNA-binding domain. However, their nuclear function is still unclear.\n\nSmad proteins have been implicated as downstream effectors of TGF beta/BMP signaling. Smad1, Smad5, and Smad8 are effectors of BMP2 and BMP4 function while Smad2 and Smad3 are involved in TGF-beta and activin-mediated growth modulation. Smad4 has been shown to mediate all of the above activities through interaction with various Smad family members. Smad6 and Smad7 regulate the response to activin/TGF beta signaling by interfering with TGF beta-mediated phosphorylation of other Smad family members.\n\nApplications:\nSuitable for use in Western Blot. Other applications have not been tested.\n\nRecommended Dilution:\nWestern Blot: 1-2ug/ml. \nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
