Streptolysin O (SLO, ASLO)

Streptolysin O from Streptococcus pyogenes, gamma-irradiated. S7974-99R is a single polypeptide chain that is not glycosylated. It has a molecular mass of ~69kD and an isoelectric point (pI) of 6.0-6.4.1. Streptolysin-O (SLO) is a bacterial protein that forms an alpha helix and is toxic to eukaryotic cells. Its toxic effect is due to the protein's ability to bind to cholesterol and form holes in the plasma membrane of cells. This ability can be utilized in the laboratory to create holes in the plasma membrane but not other membranes.\n\nUnit Definition: \nOne unit will cause 50% lysis of 50ul of a 2% human red blood cell suspension in PBS, pH 7.4, at 37 degrees C for 30 minutes.\n\nProtein Content (3-9%): As reported\n\nVial Content:\nAs reported\n\nActivity:\nAs reported\nA SLO solution will lose ~50% activity within 10 days when stored at 4 degrees C. It is recommended to freeze solutions at a minimal concentration of 0.2mg/ml.\n\nReducing Agents:\nThe activity is significantly increased in the presence of reducing agents, 20mM cysteine or 10mM dithiothreitol.\n\nStorage and Stability:\nLyophilized powder stored dessicated at -20 degrees C. Stable for 12 months. After reconstitution, aliquot and store at -20 degrees C. SLO is readily oxidized in solution.

Supplied as a powder lyophilized from a solution containing 10mM Tris-Cl buffer, 3mM sodium azide, 5mM sodium EDTA, and 1mM PMSF. Reconstitute by adding cold water (1mg/ml) into the vial and gently rotating.
