T-Complex Protein 1 alpha (TCP-1a, CCT)

TCP-1alpha is a 60kD subunit of a cytosolic hetero-oligomer chaperone (1,4) that is known to be involved in the folding of actin and tubulin. This protein is a member of the chaperonin family, which includes Escherichia coli GroEL, the mitochondrial heat-shock protein Hsp60, the plastid Rubisco-subunit-binding protein and the archaebacterial protein TF55. These chaperonins assist the folding of proteins upon ATP hydrolysis. Nine different subunits of TCP-1 containing chaperonin complexes from mammalian testis and seven different subunits of mouse F9 cells have been identified. TCP-1 has also been referred to as

This affinity purified antibody recognizes an epitope in the C-terminus of mouse TCP-1a (465-AKLRA) and will detect an ~60kD protein, corresponding to the apparent molecular mass of TCP-1alpha on SDS-PAGE immunoblots, in samples from human, mouse, rat, bovine, canine, guinea pig, hamster, monkey, porcine and rabbit origins, as well as Saccharomyces cerevisiae (weakly, consistent with the epitope sequence being AKLRS), Drosophila, Schizosaccharomyces pombe and C. elegans. In C. elegans CTA-191 reacts with TCP-1a and another CCT subunit. In plants, CTA-191 recognizes TCP-1 of Pisum sativum and the sequence of Arabidopsis thalania TCP-1a over the region of the epitope AKLRA. It has also been shown that CTA-191 reacts with a subunit of a specialized chaperonin which folds phytochrome. Cross reactivity with human Hsp60 has been \nobserved with this antibody in immunoblot analysis.
