TARDBP (TAR DNA Binding Protein, ALS10, TDP-43, OTTHUMP00000002171, TAR DNA-binding protein-43)

TARDBP was identified as the disease accumulating protein in patients with frontotemporal lobar degeneration (FTLD) with ubiquitin inclusions (FTLD-U) and in amyotrophic lateral sclerosis (ALS). TARDBP functions as a DNA-binding protein and specifically binds to the TAR DNA sequence motifs of HIV. Via this association with TAR motifs, TARDBP acts as a transcriptional repressor and inhibits HIV-1 transcription. In addition, this protein regulates alternate splicing of the cystic fibrosis transmembrane receptor (CFTR) gene.\n\nApplications: \nSuitable for use in ELISA and Western Blot. Other applications not tested.\n\nRecommended Dilution:\nPeptide ELISA: 1:16,000 \nWestern Blot: 0.3-1ug/ml Detects ~45kD band in human Jurkat cell lysates.\nOptimal dilutions to be determined by the researcher.\n\nPeptide Blocking:\nT1019-42-P: TARDBP, corresponding peptide; Peptide is supplied as a 100ul pellet. When peptide is reconstituted in 200ul water, the concentration is 0.5mg/ml. Mix equal volumes of peptide and antibody at the required dilution and leave at ambient temperature. It is best is to have two identical blots to be incubated with equal amount of antibody, but one with the antibody pre-adsorbed to the peptide for 20 minutes. Then incubate and develop the two blots in parallel.\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
