TCR, V beta 3 (T Cell Receptor) (Biotin)

Human T cell receptor (TCR) is a T cell surface expressed heterodimer of a/b or g/d chains. TCR's recognize antigen peptides bound to MHC molecules forming the basis of antigen specific response by T cells. \n\nApplications: \nSuitable for use in Flow Cytometry. Other applications not tested.\n\nRecommended Dilution:\nFlow Cytometry: 5ug/ml Labels 5x10e5 cells. CH7C17 tumor cells (HA1.7 Vb3+ transfectant)\n\nOptimal dilutions to be determined by the researcher.\n\nQuality Control:\nCH7C17 tumor cells (HA1.7 Vb3+ transfectant) were washed and incubated 45 minutes on ice with 80ul of T2040-17A TCR Vb3 at 5ug/ml. Cells were washed twice and incubated with streptavidin/R-PE, after which they were washed three times, fixed and analyzed using a BD FACstar plus. Cells stained positive with a mean shift of 1.83 log10 fluorescent units when compared to a mouse IgG2a/biotin negative control at a similar concentration. Binding was 50% blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml T2040-17 TCR Vb3 antibody.\n\nStorage and Stability:\nFor long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional Info

Specific for the human Vb3 TCR gene product and reacts with 1-10% of T cells in normal human blood. Antibody is mitogenic for T cells expressing TCR V b3.
