Tetranectin (TNA, TN, Plasminogen-kringle 4 Binding Protein)

Tetranectin (TN) is a serum and tissue protein, a C-type lectin, which binds to Ca++. It is a homotrimer of monomers each with a mass of 20kD, plasma or serum concentrations of TN are found to be approximately 10mg/l (1,2,4). In vitro, TN can bind to kringle 4 of plasminogen and enhance the activation of plaminogen to plasmin, catalyzed by tissue plasminogen activator in the presence of poly-D-lysine (3). TN is best known as a prognostic marker in ovarian cancer.\n\nApplications:\nSuitable for use in ELISA and Western Blot. Not suitable for use in Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nELISA: 1:1000 \nOptimal dilutions to be determined by the researcher.\n\nMatched Pairs:\nCapture: T2980-03\nDectection: T2980-04\n\nHybridoma: \nX63-Ag8.653 Female CF1x BALB/c mice immunized i.p. with immunogen adsorbed onto Al(OH)3. \n\nCulture Medium: \nRPMI 1640 with 10% fetal calf serum\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional Info

Recognizes amino acids 17-181 of human tetranectin monomer. Epitope specificity is shared with clones 6F192 and 6F193 as determined by inhibition ELISA. Reacts strongly with tetranectin and in sandwich ELISA in combination with a polyclonal antibody against tetranectin. In Western Blot reacts strongly with TN monomer and slightly with TN trimer. In fresh frozen tissues of ovarian cancer shows no staining for TN and no staining of paraffin-embeded, microwave-treated tissues. Especially good as capture antibody in combination with clone 6F192 in sandwich ELISA.
