Thomsen Friedenreich Antigen

The Thomsen-Fridenreich antigen acts as an oncofetal antigen, with low expression in normal adult tissues but increasing to fetal levels of expression in hyperplasia or malignancy. It is considered as a pan-carcinoma marker. Can be applied for the detection of cells with TF-antigen and is especially applicable for sensitive determination of neuraminidases.\n\nApplications:\nSuitable for use in Immunocytochemistry, Immunoprecipitation, Western Blot, Immunofluorescence and Immunohistochemistry. Other applications not tested. \n\nRecommended Dilutions: \nImmunocytochemistry: 1:10-1:50\nImmunoprecipitation: 1:10-1:50\nWestern Blot: 1:10-1:50\nImmunofluorescence: 1:10-1:50\nImmunohistochemistry (Frozen, paraffin): 1:10-1:50\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional Info

Recognizes the human Thomsen Friedenreich Antigen at ~155kD. Binds to asialoglycophorin but not to glycophorin. Agglutinates neuraminidase-treated but not untreated human erythrocytes. Positive reactions with primary breast cancer, colorectal cancer and metastases derived therefrom. Normal colonic adjacent tissue is negative. Breast epithelium (luminal membranes) may react. Positive staining with acute myelogeneous leukemia cells (HL-60, KG-1). Specific binding with both anomers of TF as well as with the TFb-related asialo-GM1.
