Cytosolic thymidine kinase 1, TK1, is a well known cell-cycle-regulated enzyme of importance in \nnucleotide metabolism as well as an activator of antiviral and anticancer drugs (1).TK1 catalyzes the ATP-dependent phosphorylation of thymidine to thymidine 5'- monophosphate, which is subsequently converted to thymidine 5'-triphosphate and utilized for DNA synthesis.In \nterminally differentiating cells, such as cardiac muscle cells, the cessation of DNA synthesis and withdrawal from the cell cycle are accompanied by the loss of TK1 activity (2).TK1 is a key enzyme of the salvage pathway for pyrimidine deoxyribonucleotide synthesis in a variety of organisms (3).Two forms have been identified in animal cells, one in cytosol and one in mitochondria.Activity of the cytosolic enzyme is high in proliferating cells and peaks during the S-phase of the cell cycle; it is very low in resting cells (4). \n \nApplications:\nSuitable for use in Flow Cytometry, Western Blot, Immunoprecipitation, Immunohistochemistry and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilution:\nFlow Cytometry: 1:70 \nWestern Blot: 1:5000-1:50,000 \nImmunoprecipitation: 1:10\nImmunohistochemistry: 1:100-1:250 \nImmunocytochemistry: 1:100\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
