Thyroid Microsomal Protein, Human (TMA, TPO, Thyroid Peroxidase)

The thyroid microsomal antigen has been shown to be the enzyme thyroid peroxidase (TPO). TPO is a membrane-bound, glycosylated, hemoprotein enzyme with a molecular weight of approximately 110,000 daltons. It plays a significant role in the biosynthesis of thyroid hormones by catalyzing both the iodination of tyrosyl residues in thyroglobulin and the coupling of the iodotyrosyl residues to form thyroxine (T4) and triiodothyronine (T3). Autoantibodies produced against TPO are capable of inhibiting the enzyme activity. They are also complement fixing antibodies that can induce cytotoxic changes in cells and are, therefore, a potential cause of thyroid dysfunction. \n\nThe measurement of low levels of TPO antibodies in serum can be useful in the assessment of a number of thyroid disorders. More than 90% of patients with autoimmune thyroiditis (Hashimoto's thyroiditis) have thyroglobulin or TPO antibodies. While not diagnostic, the detection of TPO antibodies can aid in predicting the progression of chronic thyroiditis and in further substantiating thyroid disease in patients with non-thyroidal illness. Antibodies to TPO have also been found in most patients with idiopathic hypothyroidism (85%) and Graves' disease (50%), and less frequently in patients with other thyroid disorders. Low titers may also be found in 5-10% of normal individuals. \n\nApplications: \nSuitable for use in ELISA or for antigen applications in immunological protocols. Other applications not tested. \n\nRecommended Dilution:\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C or colder. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
