TIMP1 (Tissue Inhibitor of Metalloproteinases 1, TIMP-1, TIMP, Metalloproteinase Inhibitor 1, Collagenase inhibitor, CLGI, Erythroid-potentiating Activity, EPA, Fibroblast Collagenase Inhibitor)

Complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them by binding to their catalytic zinc cofactor. Also mediates erythropoiesis in vitro; but, unlike IL-3, it is species-specific, stimulating the growth and differentiation of only human and murine erythroid progenitors. Known to act on MMP-1, MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-10, MMP-11, MMP-12, MMP-13 and MMP-16. Does not act on MMP-14.\n\nApplications:\nSuitable for use in ELISA, Western Blot, Immunohistochemistry and Neutralization. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: To detect hTIMP-1 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hTIMP-1 is 1.5-3ng/lane, under either reducing or non-reducing conditions.\nELISA: To detect hTIMP-1 by sandwich ELISA (using 100ul/well antibody solution) a concentration of 0.5-2ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human TIMP-1 as a detection antibody, allows the detection of at least 0.2-0.4ng/well of recombinant hTIMP-1.\nImmunohistochemistry: This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive ductal carcinoma. The recommended concentration is 0.1ug/ml with an overnight incubation at 4 degrees C. An HRP-labeled polymer detection system was used with a DAB chromogen. Optimal results were achieved with a proteinase K antigen retrieval.\nNeutralization: To yield one-half maximal inhibition of the biological activity of Human TIMP-1 (3ug/ml), a concentration of 5ug/ml of this antibody is required.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile dH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
