Tissue Inhibitor of Metalloproteinases 1 (TIMP1, Erythroid Potentiating Activity, EPA, Collagenase Inhibitor, CI) (BSA & Azide Free)

Tissue inhibitor of metalloproteinases-1 (TIMP-1) and Tissue inhibitor of metalloproteinases-2 (TIMP-2) are inhibitory enzymes of matrix metalloproteinase family, and are thought to be of great importance in the maintenance of connective tissue integrity. TIMP-1 forms a complex of 1:1 stoichiometry with activated interstitial collagenase, activated stromelysin, active form of 72kD Type IV collagenase (also known as MMP-2 or gelatinase A), and latent and active forms 92kD Type IV collagenase (also known as MMP-9 or gelatinase B). TIMPs inhibit the proteolytic invasiveness of tumor cells and normal placental trophoblast cells. \n\nApplications:\nSuitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested.\n\nRecommended Dilution: \nWestern Blot: 1-2ug/ml for 2hrs at RT\nImmunohistochemistry (Formalin/paraffin): 1-2ug/ml for 30 min at RT. Staining of formalin-fixed tissues is improved by boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nConditioned, serum-free medium from (TPA-treated) human fetal lung cell line HFL-1. Bladder, breast, and ovarian carcinoma. \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
