Tlk 1 (Tousled-like kinase 1)

In the plant Arabidopsis thaliana, the Tousled (Tsl) gene encodes a serine/threonine kinase that may be involved in pathways regulating cell division during the patterning of plant organs. The human Tlks share almost 50% with Arabidopsis Tsl. Both plant Tsl and Tlks localize to the nucleus, have a broad expression and display a similar substrate specificity. In\naddition, they both autophosphorylate and dimerize. Most interestingly, human Tlks are cell-cycle-regulated kinase with maximal activities linked to ongoing DNA replication. Tlk1 has a rather uniform expression level. Tlk2 is highly expressed in testis, with lower levels seen in heart, brain, liver and other organs.\n\nApplications: \nSuitable for use in ELISA, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nELISA: 0.1-1.0ug/ml\nImmunohistochemistry(1): 2ug/ml\nImmunoprecipitation(1): 5ug/IP reaction\n(native and IP-kinase assay)\nWestern Blot(1): 1-2ug/ml\n\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Specific for the ~85kD Tlk1 (Tousled-like kinase 1) protein. It does not crossreact with the related Tlk2 protein. Antibody specificity was confirmed by Western blot with epitope-tagged Tlk1 and Tlk2 proteins, Western blot and immunoprecipitation with HeLa cell lysates and by immunoprecipitation with in vitro transcribed and translated Tlk1 and Tlk2. A nuclear localization of this protein is observed by immunohistochemistry with HeLa cells. Species Crossreactivity: human.
