The Toll-like receptor (TLR) family in mammal comprises a family of transmembrane proteins characterized by multiple copies of leucine rich repeats in the extracellular domain and IL-1 receptor motif in the cytoplasmic domain. Like its counterparts in Drosophila, TLRs signal through adaptor molecules (1) and could constitute an important and unrecognized component of innate immunity in humans. The TLR family is a phylogenetically conserved mediator of innate immunity that is essential for microbial recognition (2). Ten human homologs of TLRs (TLR1-10) have been described (3). TLR3 has a restricted expression pattern being expressed in dendritic cells (DC) and lymph nodes (4,5). The expression of TLR3 in a single cell type may indicate a specific role for this molecule in a restricted setting. TLR3 is activated by dsRNA, the genetic material of some viruses, and a viral replication intermediate in others (6). Crystal struture and mutational analyses of TLR3-ectodomain suggest that Histidine-539 and Asparagine-541 are important aa residues in the recognition of dsRNA. H539E and N541A mutations abolish the dsRNA binding and ligand-dependent activation of TLR3 (7). This polyclonal antibody may block dsRNA binding to TLR3. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1-2ug/ml Immunohistochemistry (formalin fixed paraffin embedded): 5ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
