TopBP1 (DNA Topoisomerase II Binding Protein I). a phosphoprotein upregulated during the S phase of the cell cycle, which shares sequence homology with Dpb11 (S.c erevisiae), Rad4 (S. pombe), Mus101 (D. melanogaster) and Cut5 (Xenopus).Studies indicate a role for TopBP1 in DNA replication through interaction with DNA polymerase E, in S phase DNA checkpoint control, and in DNA repair response to ionizing radiation damage. TopBP1 co-localizes with BRCA1 and is an important factor in minimising stalled replication forks and double-strand breaks (DSB), thereby maintaining genomic integrity.
Applications:
Suitable for use in Immunoprecipitation and Western Blot. Other applications not tested.
Recommended Dilution:
Immunoprecipitation: 0.5-2.5ug/mg lysate
Western Blot: 1:1000-1:10,000 detects a band of ~200kD in HeLa whole cell lysates.
Optimal dilution to be determined by researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add 40-50% glycerol, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
