Topo II-a plays important roles in synthesis and transcription of DNA as well as chromosomal segregation during mitosis. It is reported to be a sensitive and specific marker of late S-, G2- & M-phases in transformed and developmentally regulated normal cells. Note that Ki67/M1B1 is also present in G1-phase. Topo II-a is also implicated in drug resistance of tumor cells.
Applications:
Suitable for use in Immunoprecipitation, Western Blot and Immunofluorescence. Other applications not tested.
Recommended Dilution:
Western Blot: 5-10ug/ml for 2h at RT
Immunoprecipitation: 10ug/mg protein lysate
Optimal dilutions to be determined by the researcher.
Positive Control:
Raji cells or exponentially growing any cultured human cells
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
