DNA topoisomerase II alpha (Topo IIa) is a 170kDa ubiquitous nuclear enzyme belonging to the topo family, which regulates the topological states of DNA. Topo II is required in chromatin condensation and segregation during mitosis. Topo II a is cell cycle regulated and its level peaks between G2 to M phase. It has been linked to cell proliferation and it may be the main isoform of Topo II involved mitotic processes (1). Topo II a passes one strand of DBA through a reversible break in a second DNA strand, which catalyzes the topological isomerisation of DNA during cell cycle (2). Topo II a overexpression has been linked to a number of human maligenacies and is the target for many chemotherapeutic agents. The majority of anticancer drugs targeting Topo IIa initiate apoptosis by stabilizing the convalent complex formed between DNA and Topo IIa (3).
Applications:
Suitable for use in Western Blot, Immunohistochemistry, Immunocytochemistry. Other applications not tested.
Recommended Dilution:
Western Blot: 1:1000
Immunohistochemistry: 1:100-1:250
Immunocytochemistry: 1:50- 1:100
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
