Transforming Growth Factor beta (TGFb)

Transforming growth factor-beta (TGFb) family of proteins has potent hematopoietic regulatory properties ranging from effects on the growth and differentiation of primitive stem cells to the differentiated functions of mature cells. Although most reports have described the inhibitory activities of TGF-beta on hematopoiesis, recent evidence supports the concept that TGF-beta can have both inhibitory and stimulatory actions on these systems. These differences depend on the differentiation state of the target cell and the other cytokines interacting with the cell. Furthermore, TGF-beta has direct bidirectional effects on cell surface expression of many cytokine receptors suggesting that it is part of the mechanism of action of TGF-beta.\n\nApplications: \nSuitable for use in ELISA, Western Blot, Neutralization and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 50-500ng/ml will allow visualization of 100-500ng/lane of TGFb\nNeutralization: Neutralizes TGFb activity in vitro in inhibition assay of CCL/64 cell growth. Also neutralizes the growth promoting action of TGFb in the NRK-49F colony forming assays. The effect of microinjection of T8250-12C into one blastomere of two cell stage Xenopus embryos indicated that T8250-12C was able to neutralize effectively the bioactivity of TGFb in vivo.\nOptimal dilution determined by the researcher.\n\nHybridoma: \nSp2/0-Ag14 myeloma cells with spleen cells from Balb/c mice. \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes human TGFb. Reacts with human platelet-derived and recombinant TGF-b in ELISA. In Western Blot, reacts with the dimeric (25kD) or the monomeric (12.5kD) molecules of TGF-b under non-reducing and reducing condition, respectively.
