Tribbles homolog 3 (Drosophila), TRIB3, is a putative protein kinase that has been implicated in the regulation of several biological processes in mammalian cells through its ability to influence Akt and other signaling pathways. It is an important negative regulator of adipogenesis because it acts at an early step in the differentiation cascade to block the C/EBPb proadipogenic function. TRB3 disrupts insulin signaling by binding directly to Akt and blocking activation of the kinase, thus contributing to insulin resistance in individuals with susceptibility to type II diabetes. TRIB3 expression is remarkably reduced in prostate cancer PC-3 cells after inhibition of phosphatidylinositol (PI) 3-kinase. The expression of TRIB3 is controlled by nutrient supplies, where the lack of glucose or amino acids results in a substantial increase in TRIB3 protein levels in a PI 3-kinase-dependent manner. Storage and Stability:
May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
