TRIM5a, otherwise known as RNF88 (RING finger protein 88), a cytoplasmic protein and member of the TRIM/RBCC family, known to play a vital role in species-specific early defence against retrovirus infection.Studies suggest that it is the association of TRIM5 with specific retroviral capsid sequences which determines the effectiveness of the defence against infection. For example, in Old World monkeys, TRIM5alpha specifically associates with HIV type 1 (HIV-1), whereas in humans TRIM5alpha is not effective against HIV-1, but can inhibit strains of the murine leukemia virus (MLV).
Applications:
Suitable for use in Immunohistochemistry (paraffin), ELISA and Western Blot. Other applications no tested.
Recommended Dilution:
Immunohistochemistry (paraffin): 1ug/ml, requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose.
ELISA: 1:16,000
Western Blot: 1-3ug/ml detects a band of a~37kD in human brain cell lysates.
Optimal dilution to be determined by researcher.
Positive Control:
Human tonsil
Storage and Stability:
May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add 40-50% glycerol, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
