Trypsin-3 (PRSS3, Mesotrypsin)

Trypsin-3, also known as mesotrypsin, is one of the three trypsins produced by the pancreas. It consists of a pro peptide (aa 16 to 23) and a mature chain (aa 24 to 247). \n \nApplications: \nSuitable for use in Neutralization and Western Blot. Other applications have not been tested. \n\nRecommended Dilutions: \nNeutralization:The antibody was preincubated with the enzyme at different molar ratios at RT for 30 minutes. The enzyme was then assayed with a fluorogenic peptide substrate. 50% of the proteolytic activity was inhibited by the antibody at approximately 0.15ug/ml(IC 50) under conditions in which the enzyme was present at 0.03ug/ml and the substrate concentration was 10uM. Considering the molecular masses of the enzyme (26kD) and the antibody (150kD), IC 50 was achieved at approximately 1:1 molar ratio of the antibody to the enzyme. \n\nWestern blot :1-2ug/ml. Using a colorimetric detection system, the detection limit for rhTrypsin-3 is ~25 ng/lane under non-reducing conditions. Chemiluminescent detection will increase sensitivity by 5 to 50 fold. Reducing conditions are not recommended. \n\nOptimal dilutions to be determined by the researcher. \n\nHybridoma: \nMouse myeloma cells were fused with B cells obtained from a rat immunized with purified, NS0-derived, recombinant human trypsin 3 encoded by the PRSS3 gene.\n\nEndotoxin: \n< 0.1 EU per 1ug of the antibody as determined by the LAL method. I\n \nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Reconstitute with sterile buffer. Aliquot and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.\n
