Tryptase, beta1 (TPSB1, MCPT7, Mast Cell Protease 7)

Mast cells contain a number of preformed chemical mediators such as histamine, chymase, carboxypeptidase and proteolytic tryptase. A substantial quantity of tryptase is estimated to be found in mast cells of skin and lung and suggestes this enzyme plays a major role in mast cell mediated events. In vitro studies indicate tryptase can cleave C3 to form C3a anaphylatoxin, inactivate fibrinogen as a coaguable substrate for thrombin and activate latent collagenase. \n\nApplications: \nSuitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilution:\nDirect ELISA: 0.5-1ug/ml. The detection limit for recombinant murine Tryptase beta1 is ~0.5ng/well.\nWestern Blot: 0.1-0.2ug/ml. The detection limit for recombinant murine Tryptase beta1 is ~1ng/lane under non-reducing and reducing conditions.\nImmunoprecipitation: Immunoprecipitates recombinant, murine Tryptase beta1 from conditioned media of transfected NS0 cells. The recovered recombinant murine Tryptase beta1 can be detected by Western Blot analysis using a monoclonal antibody.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
