Tryptophan hydroxylase (TPH) is one of a small family of monooxygenases that use tetrahydropterine as a cofactor during the catabolism of aromatic L-amino acids. It contains catalytic and regulatory domains, as well as a short tetramerization domain. TPH is the first catalytic step in the biosynthesis of serotonin in the CNS and melatonin in the pineal gland. TPH plays an important role in the pathology of several neuropsychiatric disorders, including depression, aggression, and schizophrenia. Applications: Suitable for use in Immunofluorescence, Western Blot and Immunohistochemistry (paraffin-embedded sections). Other applications not tested. Recommended Dilution: Western Blot: 1:1,000. A 55kD band should be detected. Immunohistochemistry: 1:1,000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 3 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
