Dimers of alpha and beta tubulin bind to two GTP molecules and assemble into microtubules. The GTP bound to the beta tubulin subunit eventually hydrolyses into GDP. This GTP cycle is essential for microtubule stability, as dimers bound to GTP tend to assemble into microtubules, whereas dimers bound to GDP tend to be unstable. The tubulin beta 3 chain (also known as neuron-specific class III beta-tubulin) are found exclusively in neurons and serves as a neuronal marker. It has also been found in cancer cells and correlates with an increasing histological degree of malignancy. Applications: Suitable for use in Western Blot and Immunohistochemistry (frozen, paraffin). Other applications not tested. Recommended Dilution: Western Blot: 1:10,000 Immunohistochemistry (paraffin, frozen): 1:1,000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
