Gamma tubulin, a member of the tubulin superfamily, is a ubiquitous and highly conserved protein within the microtubule organizing centre (MTOC). Gamma tubulin is not a component of microtubules, rather it functions as the microtubule nucleator at the MOTC, is responsible for binding microtubule minus ends and mediating the link between microtubles and the centrosome. By binding to the beta tubulin subunit of the tubulin molecule, it establishes the polarity of a microtubule leaving the alpha tubulin subunit exposed at the positive end. The abundance of Gamma tubulin is less than 1% pf the level of either alpha or beta tubulin. It shares approximately 28-32% identity with alpha tubulin from various organisms and 32-36% identity with beta tubulins. The detection, localization and characterization of proteins involved in microtubule function is fundamental to the understanding of mitosis, meiosis and the microtubule cytoskeleton. Antibodies reacting specifically with gamma tubulin serve as an essential tool in the detection of the presence and role of this molecule in various cellular settings. Gamma tubulin 1 and Gamma tubulin 2 are different isoforms that are 97% homologous. Applications: Suitable for use in Immunocytochemistry. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 3 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
